• Principle: Any positive or untouched isolation can be performed using MACS Cell Separation Reagents in combination with the MultiMACS Cell24 Separator Plus. Cells can be isolated from dissociated tissues, single cell-suspensions, PBMCs, or directly from blood products. StraightFrom® MicroBeads together with the MultiMACS Cell24 Separator Plus enable the fastest cell isolation directly from whole blood, buffy coat, LRSC and Leukopak® without density gradient centrifugation.
• Separations can be performed with the Multi-24 Column Block for simultaneous isolation of up to 24 small samples, or by splitting one large volume sample. Alternatively, up to 12 LS, LD, or Whole Blood Columns can be used in combination with the Single-Column Adapter.
• Background: The MultiMACS Cell24 Separator Plus contains a permanent 24-well magnet that induces a high-gradient magnetic field within columns. The field is strong enough to retain cells labeled with even small amounts of MACS MicroBeads in the columns. After transfer of columns to the Elution Chamber of MACS Elution Station, magnetically labeled cells can be eluted from columns via a small vacuum impulse.
• The touchscreen guides the user through the complete separation process. During cell separation the instrument moves the separation column block into and out of the magnet, thus allowing convenient processing of up to 24 samples in parallel. For elution of the positive fraction, the column block or single columns in the Single-Column Adapter must be transferred to the Elution Chamber. The vacuum-driven elution station ensures gentle elution of positively selected cells for excellent purity and maximal cell viability. The vacuum impulse guarantees simultaneous elution with identical parameters for every sample in every run. Pipetting steps during cell separation procedures are either performed manually with software guidance, or can be fully automated with the MultiMACS X.

Different column options enable you to adapt to sample size, type, and separation strategy. The Multi-24 Column Block supports both positive selection and depletion. The Single-Column Adapter is compatible with LS, LD, or Whole Blood Columns. LS Columns are optimized for positive selection and depletion of strongly magnetic labeled cells. LD Columns are optimized for depletion and Whole Blood Columns for cell isolation directly from whole blood, bone marrow, and buffy coat.
